Antibody-drug conjugate DAR calculations are automatically be performed using the ADC workflow in Byos.
Setting up the analysis follows the same process as the Intact workflow with the exception of the delta mass table. The delta mod list must use the following syntax in order for the DAR to be correctly calculated.
Drug(#) must be the first mod in every delta mod line with the # indicating the number of effective warheads on the construct.
Following this syntax allows for complex mass combinations while maintaining a proper DAR calculation. This table can be quickly setup using the 'Add Combinations' feature.
Using this feature you can add the name, mass, minimum, and maximum number of copies of the modification to be included in the combinations. Drug will need to be the first in the list to maintain the proper syntax. Finally, max total modifications will need to be set to a high number to ensure the full table is populated. Lastly, after hitting OK, the 0 value for any other modifications will need to be deleted from the list:
Once the project is created and the spectra deconvoluted, the software will automatically assign mass peaks, and the DAR will be calculated automatically in the Report tab.
There are two tabs in the reports specific for ADCs calculating DAR fraction and totals, and drug load distribution.
